Overview
UFOX plus cetuximab is a combination chemotherapy and targeted therapy regimen developed by Merck KGaA for the first-line treatment of metastatic colorectal cancer. The regimen integrates the UFOX backbone—comprising oral UFT (a prodrug of 5-fluorouracil consisting of tegafur and uracil), oral leucovorin (folinic acid), and intravenous oxaliplatin—with cetuximab, a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR). This combination was evaluated in the multicenter, randomized Phase II FUTURE study, where it was compared against the standard FOLFOX-4 plus cetuximab regimen. While the safety profile was found to be acceptable, the UFOX-based combination demonstrated inferior activity in terms of progression-free survival and response rates compared to the FOLFOX-4-based arm, although overall survival was comparable between the two groups.
