Overview

This is a combination chemotherapy regimen consisting of three agents: - UFT (tegafur-uracil), an oral prodrug combination that is converted to 5-fluorouracil (5-FU) in the body, acting as an antimetabolite to inhibit DNA synthesis. - Leucovorin (folinic acid), which enhances the cytotoxic effects of 5-FU by stabilizing its binding to thymidylate synthase. - Vinorelbine, a semi-synthetic vinca alkaloid that inhibits microtubule assembly, disrupting mitosis. The combination has been studied primarily for metastatic breast cancer, especially in patients previously treated with other chemotherapy regimens. The regimen offers an all-oral alternative to intravenous chemotherapy and has demonstrated tolerability with manageable toxicity profiles. The main toxicities observed include asthenia and nausea at higher dose levels. Stable disease responses have been reported in clinical studies[1][2][3][4].
