Overview

UK-369,003MR is a modified-release (MR) formulation of UK-369,003, a small molecule phosphodiesterase type 5 (PDE5) inhibitor developed by Pfizer. It was primarily investigated for the treatment of pulmonary hypertension associated with chronic obstructive pulmonary disease (COPD). By inhibiting the PDE5 enzyme, the drug prevents the breakdown of cyclic guanosine monophosphate (cGMP), which leads to smooth muscle relaxation and vasodilation in the pulmonary arteries. This mechanism is intended to lower pulmonary arterial pressure and improve exercise capacity in patients. A Phase 2 clinical trial (NCT00163098) was conducted to evaluate subject-optimized dosing, but the compound did not progress to regulatory approval.
