Ultrasmall superparamagnetic iron oxide-cyclo(Cys-Asn-Asn-Ser-Lys-Ser-His-Thr-Cys) (USPIO-R832, P03011) is a peptide-based magnetic resonance imaging (MRI) contrast agent targeted at vascular cell adhesion molecule-1 (VCAM-1). It contains an iron oxide core coated with a peptide ligand—cyclo(Cys-Asn-Asn-Ser-Lys-Ser-His-Thr-Cys)—with high affinity for VCAM-1, which is upregulated on endothelial cells during vascular inflammation and atherosclerosis. P03011 is used as a molecular imaging probe to noninvasively detect VCAM-1 expression in animal models of atherosclerosis and cerebral ischemia. In preclinical studies, intravenous injection of P03011 enables sensitive MRI detection of inflamed vascular regions by increasing signal at sites of VCAM-1 overexpression, based on specific nanoparticle accumulation at the target site.[1]
