Drug intelligence / Profile preview
Oulexin (umeclidinium bromide / vilanterol trifenatate) is an inhaled fixed-dose combination of a long-acting muscarinic antagonist (LAMA) and a long-acting beta2-adrenergic agonist (LABA). Developed by GlaxoSmithKline (GSK) in collaboration with Innoviva (formerly Theravance), it is marketed globally under the brand name Anoro Ellipta and in China as Oulexin. The combination is indicated for the long-term, once-daily maintenance treatment of airflow obstruction in patients with chronic obstructive pulmonary disease (COPD), including chronic bronchitis and emphysema. Umeclidinium bromide acts as an anticholinergic by competitively blocking muscarinic receptors (primarily M3) on airway smooth muscle, preventing acetylcholine-induced bronchoconstriction. Vilanterol trifenatate selectively stimulates beta2-adrenergic receptors, increasing intracellular cyclic AMP and promoting airway smooth muscle relaxation. Together, these complementary mechanisms provide sustained 24-hour bronchodilation to improve lung function and reduce exacerbations.
