Drug intelligence / Profile preview
Upacicalcet (also known as upasicalcet) is an intravenous small-molecule calcimimetic that acts as a positive allosteric modulator of the calcium-sensing receptor (CaSR). It was originally created by Ajinomoto Pharmaceuticals (now EA Pharma) and developed by Sanwa Kagaku Kenkyusho in Japan, where it was approved in June 2021 under the brand name Upasita for the treatment of secondary hyperparathyroidism (SHPT) in patients undergoing hemodialysis. Pathalys Pharma, in collaboration with Launch Therapeutics, is developing upacicalcet globally outside of Japan and Asia, with Phase III clinical trials completed in the United States and Europe. By directly binding to CaSR on parathyroid cells, upacicalcet suppresses the excessive secretion of parathyroid hormone (PTH) and lowers blood PTH levels, while demonstrating a lower risk of gastrointestinal side effects and hypocalcemia compared to oral calcimimetics.
