Overview

Urinary-derived human chorionic gonadotropin + epidermal growth factor (uhCG/EGF) is a biologic combination therapy derived from human pregnancy urine. While human chorionic gonadotropin (hCG) is the primary component (often associated with the brand Pregnyl), the urinary-derived preparation naturally contains epidermal growth factor (EGF). The combination is being repurposed for the treatment and prophylaxis of acute graft-versus-host disease (aGVHD) following allogeneic hematopoietic cell transplantation. Its mechanism of action is dual: hCG acts as an immunomodulator to induce immune tolerance (mimicking the maternal-fetal environment), while EGF promotes the repair of the intestinal epithelial barrier by activating the EGFR pathway. The program has received Orphan Drug Designation from the FDA and has been evaluated in Phase I and Phase II clinical trials.
