**UV1 + nivolumab + ipilimumab** is a combination regimen comprising UV1, a peptide-based telomerase cancer vaccine, with the immune checkpoint inhibitors nivolumab (a PD-1 inhibitor) and ipilimumab (a CTLA-4 inhibitor). This combination is being developed as an immunotherapeutic approach for malignancies such as unresectable malignant pleural mesothelioma. UV1 is designed to enhance anti-tumor T cell responses by inducing specific CD4+ T cell immunity against telomerase (hTERT) presented by tumors. Nivolumab and ipilimumab further enhance anti-tumor immunity by respectively blocking PD-1 and CTLA-4 pathways, which are critical negative regulators of immune activation. Data from the randomized phase 2 NIPU trial demonstrate clinical activity and improved survival outcomes for this combination in malignant pleural mesothelioma. The combination is currently under investigation in multiple ongoing phase 2 studies for additional cancers[1][2][3][4][5].
