Overview

V180 + Alhydrogel is an investigational combination vaccine comprising the tetravalent recombinant subunit dengue vaccine V180 formulated with the aluminum hydroxide adjuvant Alhydrogel. V180 consists of recombinant envelope glycoproteins (DEN-80E, the N-terminal 80% of the envelope protein) for each of the four dengue virus serotypes (DENV1–4). Alhydrogel, an aluminum-based adjuvant, is added to enhance the immune response. The vaccine is developed as a prophylactic approach for dengue infection, aiming to induce neutralizing antibodies to all four dengue serotypes. Trials indicate it is generally well tolerated and induces increased neutralizing antibody titers, especially in individuals previously exposed to dengue virus or live attenuated dengue vaccine. However, the immunogenicity in flavivirus-naive subjects has been modest. The addition of Alhydrogel is intended to boost humoral immunity, following its widespread use as a vaccine adjuvant in licensed protein subunit vaccines[1][5]. Primary developer is Merck[1]. The adjuvant is supplied by Brenntag Biosector, Frederikssund, Denmark[1].
