**V180 + ISCOMATRIX is an investigational recombinant tetravalent dengue vaccine formulation.** - The vaccine consists of truncated envelope proteins (DEN-80E) from all four dengue virus serotypes as antigens. - It is adjuvanted with ISCOMATRIX, a saponin-based nanoparticle adjuvant known to enhance both humoral and cellular immune responses by promoting antigen delivery and presentation and stimulating CD4+ and CD8+ T cell responses[2][3][4][6][7]. - Developed by Merck & Co., the combination was evaluated in Phase I clinical trials in flavivirus-naïve adults. Six different formulations containing the ISCOMATRIX adjuvant demonstrated robust immunogenicity, with seroconversion rates exceeding 85% for all four dengue serotypes after three doses[3][6][7]. - The addition of the ISCOMATRIX adjuvant resulted in higher frequencies of injection-site adverse events compared to aluminum-adjuvanted or unadjuvanted formulations but was generally well tolerated overall[6][7]. - The primary indication is prevention of dengue infection.
