Overview

**V941 + pembrolizumab** is a combination therapy under clinical investigation for the treatment of advanced or metastatic cancers with KRAS mutations, particularly non-small cell lung cancer, colorectal cancer, and pancreatic adenocarcinoma. V941 (also known as mRNA-5671) is an investigational mRNA-based cancer vaccine encoding mutant KRAS antigens, designed to elicit specific T cell responses against tumors harboring KRAS mutations. Pembrolizumab is a PD-1-blocking monoclonal antibody checkpoint inhibitor that enhances antitumor immune activity by disrupting the PD-1/PD-L1 pathway. The combination is intended to stimulate both innate and adaptive antitumor immunity: V941 primes and expands mutant KRAS-specific T cells, while pembrolizumab prevents inhibition of T cell activity within the tumor microenvironment.
