Overview

VAC85135 is an investigational **heterologous prime-boost neoantigen cancer-vaccine regimen** developed by Johnson & Johnson for mutation-positive myeloproliferative neoplasms. It comprises adenoviral and vaccinia viral vectors encoding polypeptides directed against mutant calreticulin and the JAK2 V617F mutation, with the intent of inducing neoantigen-specific antitumor T-cell responses against malignant hematopoietic cells. It was administered by intramuscular injection in a first-in-human Phase 1 study, including cohorts co-administered ipilimumab; the study found limited and generally delayed immunogenicity, no disease-modifying clinical responses, and concluded that further development for myeloproliferative neoplasms would not be pursued.
