VAI-VP705 + MF59 is an investigational vaccine combination consisting of a recombinant virus-like particle (VLP) vaccine and an adjuvant, developed for the prevention of parvovirus B19 infection. The vaccine component, VAI-VP705 (also known as MEDI-491), is composed of the major (VP2) and minor (VP1) capsid proteins of parvovirus B19, which self-assemble into non-infectious VLPs that mimic the native viral structure. MF59 is a squalene-based oil-in-water emulsion adjuvant included to enhance the immunogenicity of the vaccine by promoting a more robust and persistent antibody response. The development of this candidate was primarily led by the National Institute of Allergy and Infectious Diseases (NIAID) in collaboration with Vaccine Antigens Inc. (later acquired by MedImmune/AstraZeneca). Clinical evaluation focused on safety and the induction of neutralizing antibodies in healthy adults.
