VALO-D102 is an innovative oncolytic virus therapy developed by Valo Therapeutics using its proprietary PeptiCRAd (Peptide-coated Conditionally Replicating Adenovirus) platform. The technology involves coating the capsid of an oncolytic adenovirus with tumor-specific MHC-I-restricted peptides. This dual-action approach combines the direct oncolytic effect of the virus—which causes immunogenic cell death and releases danger signals—with a potent, peptide-driven vaccine effect that activates the patient's own T-cells against specific tumor antigens. By turning the tumor into an in situ vaccine, VALO-D102 aims to overcome the immunosuppressive tumor microenvironment and induce a systemic anti-tumor immune response. It is currently being investigated in clinical trials for various solid tumors, including melanoma, non-small cell lung cancer, and triple-negative breast cancer.
