Overview

**Valproate + amisulpride** is a combination of two pharmaceutical agents used primarily for the treatment of acute mania in bipolar I disorder. Valproate is a widely used mood stabilizer and anticonvulsant that primarily increases gamma-aminobutyric acid (GABA) levels in the brain, providing mood-stabilizing and antimanic effects. Amisulpride is an atypical antipsychotic that selectively antagonizes dopamine D2 and D3 receptors, particularly in the limbic system, reducing both positive and negative psychotic symptoms and extending some antidepressant effects. Clinical studies indicate that the combination is as effective as valproate with typical antipsychotics, but with a lower rate of extrapyramidal side effects and treatment-emergent adverse events[1][2][3]. This combination is most studied as an acute therapy in adults with manic episodes associated with bipolar I disorder.
