Overview

Valsacombi is a fixed-dose combination antihypertensive medication developed by Krka, d.d., Novo mesto. It contains two active ingredients: valsartan, an angiotensin II receptor blocker (ARB), and hydrochlorothiazide, a thiazide diuretic. Valsartan works by selectively blocking the binding of angiotensin II to the type-1 angiotensin II (AT1) receptor, which inhibits vasoconstriction and aldosterone release. Hydrochlorothiazide acts on the distal convoluted tubule of the kidney to inhibit the sodium-chloride cotransporter (NCC), leading to increased excretion of sodium and water and a subsequent reduction in plasma volume. This combination is indicated for the treatment of essential hypertension in adults whose blood pressure is not adequately controlled by monotherapy. The efficacy and safety of Valsacombi, including its effects on erectile function in male participants, were evaluated in the Phase 4 VICTORY trial conducted in Slovenia and other European countries.
