Overview
Vandetanib + anastrozole is a combination therapy investigated for the neoadjuvant treatment of postmenopausal patients with Stage I-III hormone receptor-positive (HR+) breast cancer. Vandetanib is an oral small molecule tyrosine kinase inhibitor that dual-targets vascular endothelial growth factor receptor-2 (VEGFR-2) and epidermal growth factor receptor (EGFR), aiming to inhibit angiogenesis and tumor cell proliferation. Anastrozole is a non-steroidal aromatase inhibitor that reduces circulating estrogen levels, a key driver in HR+ breast cancer. The combination was studied in a Phase 2 trial led by Stanford University (NCT00481845) to evaluate if blocking multiple signaling pathways could overcome resistance to endocrine therapy alone and improve clinical outcomes in the neoadjuvant setting.
