Overview
Vaniprevir + peginterferon alfa-2b + ribavirin is a three-drug combination regimen for the treatment of **chronic hepatitis C virus (HCV)** infection, especially genotype 1. **Vaniprevir** is a small molecule, macrocyclic NS3/4A protease inhibitor that blocks HCV replication by inhibiting viral polyprotein processing. **Peginterferon alfa-2b** is a pegylated form of interferon alfa-2b, acting as an immunomodulator by stimulating antiviral responses. **Ribavirin** is a nucleoside analog antiviral agent that enhances interferon effects and disrupts viral RNA synthesis. This combination enhances antiviral efficacy compared to dual therapy and is approved/was investigated primarily for use in Japanese or Asian populations. Efficacy is highest in genotype 1 infection, with significant sustained virologic response (SVR) rates in both treatment-naive and treatment-experienced patients. The most frequent adverse events are mild/moderate and include pyrexia, GI symptoms, hematologic changes, and dermatologic effects.
