Vantictumab is a fully human IgG2 monoclonal antibody that targets and binds to Frizzled (FZD) receptors 1, 2, 5, 7, and 8, thereby inhibiting canonical WNT signaling. This pathway is implicated in the maintenance of cancer stem cells and tumor growth. Paclitaxel is a well-established chemotherapeutic agent that stabilizes microtubules and inhibits cell division. The combination of vantictumab with paclitaxel has been investigated in phase Ib clinical trials for HER2-negative breast cancer and metastatic pancreatic adenocarcinoma. Preclinical studies suggest that vantictumab can sensitize cancer stem cells to paclitaxel, resulting in enhanced tumor growth inhibition compared to either agent alone. However, development was limited by bone-related toxicities such as fragility fractures attributed to WNT pathway inhibition[1][2][6].
