Varseta-M (varsetatug masetecan, CX-2051) is a conditionally activated antibody-drug conjugate (ADC) targeting epithelial cell adhesion molecule (EpCAM). It utilizes CytomX's proprietary Probody technology, which masks the antibody with a peptide to prevent binding to healthy tissue until it is cleaved by tumor-associated proteases in the tumor microenvironment. The ADC is armed with masetecan, a topoisomerase-1 inhibitor payload. This combination therapy evaluates Varseta-M in conjunction with the VEGF inhibitor bevacizumab and standard-of-care cytotoxic chemotherapy for the second-line (2L) treatment of metastatic colorectal cancer (mCRC). The regimen aims to leverage the targeted delivery of the Topo-1 payload alongside anti-angiogenic and cytotoxic effects to overcome resistance in EpCAM-expressing colorectal tumors.
