VBIR-2 + sasanlimab + tremelimumab is a combination immunotherapy regimen that combines a vaccine prime/boost strategy with immune checkpoint inhibitors. The regimen consists of three components: 1. VBIR-2: A vaccine-based immunotherapy that uses a two-part approach with an adenovirus vaccine prime followed by plasmid DNA boost to deliver tumor-associated antigens to stimulate the immune system. 2. Sasanlimab (PF-06801591): A humanized anti-PD-1 antibody that blocks the interaction between PD-1 and its ligands PD-L1 and PD-L2, preventing immune suppression and enhancing T-cell activity against cancer cells. 3. Tremelimumab: An anti-CTLA-4 antagonist antibody that blocks immune checkpoints, further enhancing the immune response against cancer cells. This combination is designed to stimulate strong CD8 and CD4 T cell responses against tumor antigens while simultaneously removing immunosuppressive mechanisms used by cancer cells.
