Overview

VBIR-2 + tremelimumab is a combination cancer immunotherapy regimen that includes a vaccine-based immunotherapy (VBIR-2) combined with the immune checkpoint inhibitor tremelimumab. The vaccine component uses tumor-associated antigens delivered via a priming vaccination with a chimpanzee adenovirus vector and booster vaccinations with electroporated DNA to stimulate strong antigen-specific CD4 and CD8 T cell responses. Tremelimumab is an anti-CTLA-4 monoclonal antibody that acts as an immune checkpoint inhibitor, blocking CTLA-4 to enhance T cell activation and anti-tumor immunity. This combination aims to break immune tolerance against tumor antigens, boost the body's immune response against cancer cells, and improve clinical outcomes in cancers such as metastatic castration-resistant prostate cancer (mCRPC), non-small cell lung cancer (NSCLC), and triple-negative breast cancer (TNBC). Clinical trials have shown manageable safety profiles, antigen-specific immunity stimulation, modest antitumor activity especially in androgen-sensitive prostate cancer, and some durable PSA responses without androgen deprivation therapy.
