Veliparib is a small molecule inhibitor of poly (ADP-ribose) polymerase (PARP)-1 and PARP2, developed by AbbVie. It sensitizes tumor cells to chemotherapy and radiation by inhibiting DNA repair mechanisms, particularly in tumors with homologous recombination deficiency. mFOLFOX6 is a modified chemotherapy regimen consisting of oxaliplatin, 5-fluorouracil (5-FU), and leucovorin, commonly used for colorectal cancer but also applied in other gastrointestinal malignancies. The combination of veliparib with mFOLFOX6 has been investigated primarily for metastatic pancreatic ductal adenocarcinoma (mPDAC), showing tolerability and promising activity especially in patients with HR-DDR mutations or platinum-naive disease[1][4][8].
