Venetoclax + AMG 176 is an experimental combination of two targeted small molecule drugs designed to induce apoptosis in malignant hematologic cells. Venetoclax is a selective BCL2 inhibitor, approved for the treatment of chronic lymphocytic leukemia (CLL) and certain types of acute myeloid leukemia (AML), while AMG 176 is a clinical-stage inhibitor of the anti-apoptotic protein MCL1. Together, these agents aim to simultaneously block the key anti-apoptotic members of the BCL2 family that mediate resistance to apoptosis in many blood cancers. Preclinical studies show additive or synergistic efficacy in models of CLL and AML, and early-phase clinical trials have been initiated to test safety and efficacy in human patients. Mechanistically, the combination targets both BCL2 and MCL1 pathways, critical regulators of cancer cell survival, intending to overcome resistance seen with inhibition of either pathway alone[1][3][5][6][7].
