Overview

This is a **combination regimen of four drugs**: venetoclax (a selective BCL-2 inhibitor), daratumumab (an anti-CD38 monoclonal antibody), bortezomib (a proteasome inhibitor), and dexamethasone (a synthetic glucocorticoid corticosteroid). Venetoclax induces apoptosis in myeloma cells by inhibiting BCL-2, especially effective in patients with t(11;14) translocation; daratumumab binds CD38 and mediates direct tumor cell lysis and immunomodulation; bortezomib inhibits the 26S proteasome, disrupting protein degradation in malignant plasma cells; dexamethasone acts as an anti-inflammatory and anti-cancer corticosteroid. This combination is being investigated in clinical trials for efficacy and safety in relapsed/refractory multiple myeloma, including in patients both with and without the t(11;14) translocation[2][5][9].
