Venetoclax + daunorubicin + cytarabine is a combination regimen under investigation for the treatment of acute myeloid leukemia (AML), particularly in newly diagnosed or high-risk patients. Venetoclax is a selective B-cell lymphoma 2 (BCL-2) inhibitor that promotes apoptosis in cancer cells dependent on BCL-2 for survival. Daunorubicin is an anthracycline antibiotic that intercalates DNA, inhibits topoisomerase II, and generates free radicals leading to DNA damage and cell death. Cytarabine is an antimetabolite that inhibits DNA synthesis during the S-phase of the cell cycle by acting as a pyrimidine analog. This combination aims to enhance anti-leukemic efficacy by targeting multiple pathways involved in leukemic cell survival and proliferation[1][2][5][7]. The regimen has shown promising response rates but can be associated with significant myelosuppression.
