Venetoclax + olverembatinib + prednisone, also known as the VOP regimen, is a combination therapy being evaluated for the treatment of Philadelphia chromosome-positive (Ph+) B-cell acute lymphoblastic leukemia (B-ALL). The regimen combines three distinct therapeutic agents: olverembatinib (HQP1351), a third-generation BCR-ABL tyrosine kinase inhibitor (TKI) designed to overcome resistance mutations like T315I; venetoclax, a selective BCL-2 inhibitor that promotes apoptosis in leukemic cells; and prednisone, a glucocorticoid with lymphocytotoxic properties. This combination is specifically being studied in Phase II clinical trials led by the First Affiliated Hospital of Zhejiang University to improve deep molecular response rates and survival outcomes in adult patients with newly diagnosed Ph+ B-ALL, a high-risk subtype of leukemia.
