A combination therapy consisting of venetoclax, an oral BCL-2 inhibitor, and pegcrisantaspase, a pegylated form of crisantaspase that depletes plasma glutamine. This novel regimen works through synergistic mechanisms where venetoclax induces apoptosis by inhibiting BCL-2, while pegcrisantaspase depletes plasma glutamine, disrupts mRNA translation, and decreases expression of proteins like MCL-1. The combination has shown efficacy in relapsed/refractory acute myeloid leukemia (AML), including in patients who had previously progressed on venetoclax-based therapies.
