**Vesatolimod + ART** is a combination therapy consisting of vesatolimod, a small molecule **Toll-like receptor 7 (TLR7) agonist**, and antiretroviral therapy (ART). Vesatolimod activates the TLR7 pathway, leading to upregulation of innate immune responses, interferon signaling, and immune cell activation (notably dendritic cells and natural killer cells). This has been investigated as part of HIV cure strategies—specifically as a latency-reversing agent to reactivate latent HIV-infected cells so that ART can eliminate them. Combination therapy with ART has been shown to suppress viral load quickly, delay antibody responses to HIV, reduce the HIV reservoir, and modestly delay viral rebound following ART interruption. Vesatolimod is under development for HIV functional cure regimens and is not yet approved. The developer of vesatolimod is Gilead.
