Overview

This drug is a **combination therapy** of three investigational agents with distinct, complementary mechanisms, being used predominantly in HIV cure and prevention research: - **vesatolimod** is an oral small molecule **Toll-like receptor 7 (TLR7) agonist**, which stimulates plasmacytoid dendritic cells and B cells to elicit a potent immune response, including interferon-alpha production and activation of innate and adaptive immunity. It is being evaluated as a latency-reversing agent and immune modulator[1][3][5][7][9]. - **VRC07-523LS** and **CAP256V2LS** are **broadly neutralizing monoclonal antibodies (bNAbs)**. VRC07-523LS targets the HIV-1 CD4 binding site, with an LS mutation to extend its half-life and enhance potency and breadth against HIV-1[4][6][8]. CAP256V2LS targets the V1/V2 glycan on the HIV envelope, is derived from the CAP256-VRC26.25 lineage, and features both an LS mutation for half-life extension and an engineered site to prevent proteolytic clipping while retaining high neutralizing breadth and potency against clade C viruses[2][4][8]. The combination is being evaluated for **HIV cure strategies**—aiming to reduce or eliminate the HIV reservoir (latency reversal and immune clearance), possibly in the context of antiretroviral therapy (ART) interruption or prevention.
