**VGX-3100 + imiquimod** is an investigational combination therapy for the treatment of high-grade squamous intraepithelial lesion (HSIL) of the vulva and related HPV-16 and/or HPV-18-induced precancerous lesions. VGX-3100 is a DNA-based immunotherapy designed to induce cell-mediated immune responses against the E6 and E7 oncoproteins of HPV-16 and HPV-18, delivered intramuscularly with electroporation. Imiquimod is a topical immune response modifier that acts as a toll-like receptor 7 agonist, stimulating local innate and adaptive immunity. The rationale for combining these drugs is to potentiate anti-HPV immune responses and enhance lesion regression, as studied in clinical trials (e.g., NCT03180684) for vulvar HSIL caused by HPV-16/18[1][3][5].
