This is a combination therapy consisting of VIB4920 (also known as dazodalibep or MEDI4920) and a tumor necrosis factor (TNF) inhibitor. VIB4920 is a recombinant fusion protein engineered to antagonize CD40 ligand (CD40L), thereby inhibiting the CD40/CD40L pathway, which plays a critical role in T-cell and B-cell interactions and immune activation. It is designed to reduce autoimmune activity by blocking this co-stimulatory signal without causing adverse haemostasis seen with earlier anti-CD40L agents[2][4][5]. TNF inhibitors are biologic drugs that suppress the pro-inflammatory cytokine TNF-alpha, widely used in autoimmune diseases such as rheumatoid arthritis, psoriatic arthritis, Crohn's disease, ulcerative colitis, ankylosing spondylitis, and others[6][9]. The combination aims to provide dual immunomodulation by targeting both T/B cell co-stimulation (via CD40L blockade) and inflammatory cytokine signaling (via TNF inhibition). This approach may be investigated for enhanced efficacy in treating autoimmune diseases.
