Vibecototamab + putilimab is a combination therapy being evaluated for the treatment of EGFR-positive refractory advanced biliary tract malignancies. Vibecototamab (also known as MRG003) is an antibody-drug conjugate (ADC) composed of a humanized anti-EGFR monoclonal antibody conjugated to the microtubule inhibitor monomethyl auristatin E (MMAE) via a protease-cleavable valine-citrulline linker. Putilimab (also known as HLX10 or puzelimab) is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, acting as an immune checkpoint inhibitor. The combination aims to leverage the targeted cytotoxicity of the ADC, which induces immunogenic cell death, and the immune-restoring capabilities of the PD-1 inhibitor to overcome resistance in advanced biliary tract cancers. This specific combination is being investigated in clinical trials led by West China Hospital.
