Overview

This is a combination regimen consisting of **vibostolimab** (an anti-TIGIT monoclonal antibody), **pembrolizumab** (an anti–PD-1 immune checkpoint inhibitor), **carboplatin** (a platinum-based chemotherapeutic agent), and **etoposide** (a topoisomerase II inhibitor). - **Vibostolimab** acts by blocking TIGIT, an inhibitory receptor on immune cells, potentially enhancing anti-tumor immune responses. - **Pembrolizumab** blocks PD-1 to release the inhibition of cytotoxic T cells, promoting immune-mediated tumor cell killing. - **Carboplatin** is a platinum compound causing DNA cross-linking and apoptosis in rapidly dividing cells. - **Etoposide** inhibits topoisomerase II, leading to DNA damage and cell death primarily in actively proliferating cells. The regimen is being investigated primarily for use in **extensive-stage small cell lung cancer (ES-SCLC)** and other neuroendocrine tumors. It combines two immunotherapies with standard chemotherapy to potentially improve response rates and survival over chemotherapy alone[2][3][4][5]. Reported studies feature similar regimens, though most published data are for pembrolizumab plus carboplatin and etoposide, and the addition of vibostolimab is currently under clinical investigation.
