Overview

This combination consists of **vibostolimab** (an anti–TIGIT monoclonal antibody), **pembrolizumab** (an anti–PD-1 monoclonal antibody), **cisplatin** (a platinum-based DNA crosslinking agent), and **etoposide** (a topoisomerase II inhibitor). The combination aims to enhance immune checkpoint blockade (anti–PD-1, anti–TIGIT pathways) while leveraging direct tumoricidal effects via DNA damage and inhibition of DNA repair. While pembrolizumab + cisplatin + etoposide is established in clinical trials for extensive-stage small cell lung cancer and other indications, adding vibostolimab is the subject of recent and ongoing studies, primarily led by Merck, for multiple cancer indications, including extensive-stage small cell lung cancer and other solid tumors[2].
