Drug intelligence / Profile preview
vimentin35-71Cit64 + calcitriol is an antigen-specific immunotherapy (ASI) liposomal formulation developed by the University of Queensland and its spin-off Dendright. It consists of liposomes (formulated with semi-synthetic phospholipids POPC and POPG) co-encapsulating the citrullinated autoantigen peptide vimentin35-71Cit64 and the NF-kB inhibitor calcitriol (1,25-dihydroxycholecalciferol). The therapy is designed to restore immune tolerance in patients with rheumatoid arthritis (RA). Following subcutaneous administration, the liposomes target antigen-presenting cells (APCs) in draining lymph nodes, where calcitriol inhibits NF-kB to promote a tolerogenic phenotype, and the vimentin peptide is presented to CD4+ T cells to induce regulatory T cells (Tregs) and suppress pro-inflammatory autoimmune responses.
