Dactinomycin (also known as actinomycin-D) is an antitumor antibiotic that intercalates into DNA and inhibits RNA synthesis by blocking RNA polymerase. This dual-mechanism approach has been a standard of care for over 40 years in treating pediatric cancers such as Wilms tumor (nephroblastoma) and rhabdomyosarcoma. The regimen is frequently administered as the "VA" component of more complex protocols, such as the VAC regimen (vincristine, actinomycin, and cyclophosphamide), and is typically delivered via intravenous injection.
