vincristine + doxorubicin + prednisone + asparaginase

A multi-agent chemotherapy regimen combining vincristine, doxorubicin, prednisone (or prednisolone), and asparaginase, primarily used for the treatment of adult acute lymphoblastic leukemia (ALL)[1]. Doxorubicin and vincristine are cytotoxic agents that impair DNA synthesis and microtubule assembly, respectively, leading to cell death. Prednisone is a glucocorticoid that induces apoptosis in lymphoid cells. Asparaginase is an enzyme that depletes asparagine, an amino acid critical for the survival of leukemic cells[1]. The inclusion of L-asparaginase in this regimen was studied in a randomized, multicenter trial, but did not demonstrate a statistically significant benefit in overall survival or remission rates compared to the same regimen without asparaginase[1]. Adverse effects such as pancreatitis and coagulopathy are more common with the asparaginase-containing combination[1].
