Drug intelligence / Profile preview
The combination of vincristine, etoposide, prednisone, doxorubicin, cyclophosphamide, and dacarbazine, commonly referred to as the **DECOPDAC** (or **DECOPDAC-21**) regimen, is an intensified consolidation chemotherapy regimen used in pediatric and adolescent patients with classical Hodgkin lymphoma (cHL). Developed as part of the EuroNet-PHL-C2 clinical trial, this regimen is designed to intensify chemotherapy in intermediate-risk and advanced-risk patients to reduce or eliminate the need for subsequent radiotherapy, thereby minimizing long-term toxicities such as secondary malignancies and cardiovascular issues. The components of the regimen work through distinct, complementary mechanisms: vincristine inhibits tubulin polymerization to disrupt mitosis; etoposide and doxorubicin target DNA topoisomerase II to induce double-strand breaks; prednisone acts as a glucocorticoid receptor agonist to promote apoptosis; and cyclophosphamide and dacarbazine function as DNA alkylating agents to prevent replication.
