Overview

Vincristine + topotecan is a combination chemotherapy regimen consisting of two small molecule anticancer agents, vincristine and topotecan. Vincristine is a vinca alkaloid that inhibits microtubule formation in mitotic spindle, arresting cell division at metaphase. Topotecan is a topoisomerase I inhibitor that prevents DNA unwinding and replication by stabilizing the cleavable complex between DNA and the enzyme, leading to DNA damage and apoptosis. This combination has demonstrated synergistic antitumor activity in preclinical models when administered at specific ratios[1]. Clinically, it has been used for advanced intraocular retinoblastoma[2], as well as investigated in pediatric solid tumors such as neuroblastoma, Ewing sarcoma, rhabdomyosarcoma, and osteosarcoma[3][4][5]. The combination may be delivered as free drugs or co-encapsulated in nanoliposomes to optimize pharmacokinetics and therapeutic efficacy[1].
