Drug intelligence / Profile preview
Vincristine sulfate liposome is a liposomal formulation of the vinca alkaloid vincristine, designed to enhance the drug's therapeutic index by prolonging circulation time and increasing delivery to tumor tissues. The active ingredient, vincristine sulfate, is encapsulated within sphingomyelin/cholesterol liposomes. Its mechanism of action involves binding to tubulin and inhibiting microtubule polymerization, which leads to mitotic arrest in metaphase and subsequent apoptosis of rapidly dividing cancer cells. In the clinical setting, specifically within the Children's Oncology Group (COG) trial AHOD0831, this formulation was evaluated as a component of the ABVE-PC chemotherapy regimen (doxorubicin, bleomycin, vincristine, etoposide, prednisone, cyclophosphamide) for the treatment of newly diagnosed high-risk Hodgkin lymphoma in pediatric and adolescent patients. While the branded version, Marqibo, is FDA-approved for adult Philadelphia chromosome-negative acute lymphoblastic leukemia, its application in Hodgkin lymphoma has been primarily investigated in Phase III response-adapted therapy trials.
