Vinflunine is a novel fluorinated vinca alkaloid and microtubule inhibitor developed by Pierre Fabre Médicament. It is specifically indicated as a second-line treatment for advanced or metastatic transitional cell carcinoma of the urothelial tract (TCCU) after failure of a prior platinum-containing regimen. Unlike earlier vinca alkaloids, vinflunine exhibits a unique mechanism of action characterized by weaker binding to tubulin, which translates to higher antitumour activity and a distinct safety profile. In clinical practice, it is often administered alongside best supportive care (BSC), a regimen validated in a landmark Phase III trial (NCT00315237) that demonstrated a significant overall survival benefit compared to BSC alone.
