Overview

Vinorelbine + oxaliplatin is a combination chemotherapy regimen consisting of two small molecule antineoplastic agents: vinorelbine, a semi-synthetic vinca alkaloid that inhibits microtubule assembly, and oxaliplatin, a third-generation platinum-based compound that forms DNA adducts to inhibit DNA replication and transcription. This combination has been studied primarily in advanced non-small cell lung cancer (NSCLC), metastatic breast cancer (MBC), malignant pleural mesothelioma, and triple-negative breast cancer (mTNBC). The regimen is typically administered intravenously on an outpatient basis. Vinorelbine disrupts mitosis by binding tubulin, while oxaliplatin induces apoptosis through DNA crosslinking. The combination demonstrates synergistic effects in inhibiting tumor proliferation, migration, invasion, and inducing apoptosis[1][2][6]. Common toxicities include neutropenia (sometimes severe), neurosensory toxicity from oxaliplatin (usually mild to moderate), gastrointestinal symptoms such as vomiting and diarrhea[1]. The regimen is considered active with manageable toxicity profiles in several solid tumors.
