VIP943 is a next-generation antibody-drug conjugate (ADC) developed by Vincerx Pharma for the treatment of CD123-positive hematologic malignancies, including acute myeloid leukemia (AML), B-cell acute lymphoblastic leukemia (B-ALL), and myelodysplastic syndrome (MDS). It is the first ADC derived from Vincerx's VersAptx platform. The molecule consists of a humanized anti-CD123 monoclonal antibody conjugated to a novel kinesin spindle protein inhibitor (KSPi) payload via a proprietary legumain-cleavable linker. A key feature of VIP943 is the 'CellTrapper' technology, which modifies the payload to prevent it from diffusing out of the target cancer cell once released, thereby increasing intracellular accumulation and potentially reducing systemic toxicities like neutropenia often seen with other ADCs. It is currently being evaluated in Phase 1 clinical trials for patients with relapsed or refractory CD123+ malignancies.
