Overview

This is an **investigational combination regimen** for chronic hepatitis B, comprising three agents: - **VIR-2218** (an RNA interference small interfering RNA [siRNA] designed to target hepatitis B virus RNA, reducing viral replication and antigen expression); - **Selgantolimod** (an oral agonist of Toll-like receptor 8 [TLR8], which modulates innate and adaptive immune responses by enhancing cytokine and chemokine release and activating immune cell subsets); - **Nivolumab** (a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 [PD-1], reversing T-cell exhaustion and restoring antiviral immunity). This combination is **being studied for inducing functional cure in chronic hepatitis B** through antiviral, immune stimulation, and immunomodulation approaches. Clinical trial participation includes nucleos(t)ide analogue-treated and viremic hepatitis B subjects. Development is led by Vir Biotechnology (VIR-2218), Gilead Sciences (selgantolimod), and Bristol Myers Squibb (nivolumab/Opdivo), with manufacturing by these respective companies.
