Overview

This is a combination of three drugs: vismodegib, a small molecule inhibitor of the hedgehog signaling pathway, and norethindrone plus ethinyl estradiol, which together are commonly used as combined oral contraceptives or hormone therapy. Vismodegib is primarily used as an antineoplastic agent for the treatment of basal cell carcinoma, acting to inhibit Smoothened, a component of the hedgehog pathway, thereby blocking tumor growth. Norethindrone is a synthetic progestin, and ethinyl estradiol is a synthetic estrogen; together, they function as hormonal contraceptives or hormone replacement therapy, suppressing ovulation, altering the endometrial lining, and causing changes in cervical mucus to prevent pregnancy or treat symptoms of menopause. This combination may be considered in situations where vismodegib is prescribed to patients who require continued contraception due to vismodegib’s teratogenicity, or in clinical studies examining drug-drug interactions[1][2][3][4][5].
