This is a combination regimen of four agents: - **Visugromab** is a monoclonal antibody that neutralizes Growth Differentiation Factor 15 (GDF-15), a protein implicated in resistance to immune checkpoint inhibitors in cancer, in development for overcoming resistance to anti-PD-1/PD-L1 therapy[1][7]. - **Pembrolizumab** is an anti–programmed death-1 (PD-1) immune checkpoint inhibitor, approved for first-line and subsequent treatment of various cancers, notably non-small cell lung cancer (NSCLC)[2][5][8]. - **Pemetrexed** is an antifolate antimetabolite chemotherapy that disrupts folate-dependent metabolic processes essential for cell replication, commonly used for non-squamous NSCLC and mesothelioma[9]. - **Carboplatin** is a platinum-based chemotherapy agent that induces DNA crosslinking, leading to apoptosis of rapidly dividing cells, widely used for many solid tumors[9]. This new combination is designed to leverage immune modulation (visugromab, pembrolizumab) with cytotoxic chemotherapy (pemetrexed, carboplatin) in difficult-to-treat advanced solid tumors, including metastatic non-squamous NSCLC, especially following progression on prior checkpoint inhibitors[1][7].
