This combination therapy was evaluated in a pilot clinical trial (NCT01890772) led by Dr. Timothy Morgan for the treatment of treatment-naive veterans with genotype 1 chronic hepatitis C. The regimen consists of vitamin D3 (cholecalciferol) added to the standard-of-care triple therapy at the time: telaprevir (an NS3/4A protease inhibitor), peginterferon alfa-2a (an immune modulator), and ribavirin (a nucleoside analog). Vitamin D3 was administered at 5,000 IU daily during a lead-in phase and 15,000 IU weekly during the 12-week treatment phase. The study aimed to determine if vitamin D supplementation could enhance the virologic response and safety profile of the triple therapy by modulating the host immune response and potentially improving viral clearance.
