Drug intelligence / Profile preview
VLPDEN-02 + ALFQ is an investigational tetravalent dengue virus-like particle (VLP) vaccine developed by VLP Therapeutics. The vaccine consists of virus-like particles engineered from the structural envelope (E) and pre-membrane (prM) proteins of all four dengue virus serotypes (DENV-1, DENV-2, DENV-3, and DENV-4), formulated with the Army Liposome Formulation with QS-21 (ALFQ) adjuvant. Because it lacks a viral genome, the vaccine is non-infectious but mimics the native antigenic structure of the virus to elicit broadly neutralizing antibodies. In preclinical studies in non-human primates, the vaccine induced high levels of neutralizing antibodies against all four serotypes for over a year and reduced viral replication without generating antibody-dependent enhancement (ADE) responses. It is currently being evaluated in a Phase 1 clinical trial in healthy flavivirus-naïve adults for the prevention of dengue fever and dengue hemorrhagic fever.
