VMD-928 + pembrolizumab is an investigational combination therapy being developed by VM Oncology, LLC for the treatment of TrkA-driven advanced solid tumors and lymphoma. VMD-928 is a first-in-class, oral, selective, allosteric, and irreversible inhibitor of Tropomyosin receptor kinase A (TrkA/NTRK1). Unlike traditional ATP-competitive inhibitors, VMD-928 binds to an allosteric site, which may provide superior selectivity and overcome resistance mechanisms associated with ATP-binding site mutations. Pembrolizumab is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, acting as a checkpoint inhibitor to restore T-cell mediated anti-tumor immunity. This combination aims to synergistically inhibit tumor growth and enhance the immune response in patients with tumors overexpressing TrkA protein.
